He married Aletta Johanna Sophia Glaeser on 12 Dec 1848 at Worcester, Cape, South Africa. Aletta Johanna Sophia was the daughter of George William Henry Glaeser and Dorothea Catharina Cornelia van Dyk. She was born on 24 Jul 1826.

They had 5 children: William Henry Vos, Dorothea Maria Georgina Vos, Elizabeth Johanna Vos, George Glaeser Vos and George Henry Vos.
